Protocol used
1) Induced for 1 hour at 30 degrees2) Labeled for 2 hours at 30 degrees (in presence of 1.8 micromolar ReAsH-EDT2)3) Post fixed with _0.01%_ Acrolein and 2.5% of Glut. in 0.01M Sodium Cocadalyte buffer.4) HPF5) FS by using Epoxy resin as fixative: 20% of the complete Araldite/Epon formulation in acetone. Infiltration and embedding in 100% of resin complex.6) 0.5um sections were cut and collected on gold grids. Sections were post stained with 4% of Osmium followed by Sato lead and finally 10nm was applied on both sides of sections.
