Isolated chloroplas tmembranes of the unicellular photosynthetic alga Chlamydomonas reinhardtii were fixed, sectioned, and examined by transmission electron microscopy. Under the isolation conditions used, some stacked thylakoids are formed in vitro (S) in this ac-31 mutant. Also seen are starch grains (St) and a pyrenoid (P). Isolated chloroplast membranes were fixed with glutaraldehyde, post fixed with osmium tetroxide, dehydrated, embedded in epoxy resin, and thin sections cut, stained and examined with a Hitachi HU-11C TEM.
