Description

Migration of neuronal precursors along from the subventricular zone to the olfactory bulb via the rostral migratory stream. Parasagittal slices were prepared from P10 mice (see Webb et al.,PMID: 12071004); they contained the entire RMS, which originates at the subventricular zone (SVZ) and ends at the center of the olfactory bulb (OB). The slices were cultured on a Millicell-CM membrane and small crystals of DiI were placed onto the RMS using a microneedle. A field in each slice was selected that retained good morphology and had fluorescently labeled migrating cells adjacent to the DiI crystals. Images were captured over 4 h using 0.20 s exposures. Images were acquired using a Nikon IX-70 inverted microscope fitted with a cooled CCD camera (Photometrics CH250). Fluorescence images were recorded every 5 min using a 10× objective.
