Description

Effect of depletion of the Nup107-160 nucleoporin on spindles assembled in vitro with sperm chromatin and in Xenopus oocyte extracts. Spindles are labeled with tubulin (red) and chromosomes are stained with DAPI (blue). Lower far right panel shows a rare conformation. See Orjalo et al 2006. Mol Biol Cell 17:3806-3808.

Technical Details

Xenopus egg extracts were mixed with sperm chromatin, treated to induce spindle formation, antibodies added, and the preparations fixed with paraformaldehyde, and centrifuged onto cover slips through a glycerol cushion, processed for immunofluorescence and examined using a Zeiss Axioskop microscope. See Fig 6A in Orjalo et al. 2006 Mol Biol Cell 17:3806-3818.
