Description

The AZM enters the oral cavity of Euplotes where it occupies the right side of the buccal cavity while cytopharyngeal lamellae composed of microtubules arise from the left wall of the cavity. Large vesicles that contain lipid-like osmiophilic material line up between the ribbons and make close contact with the single membrane of the food vacuole-forming cytopharynx. Under different fixation conditions these vesicles are compact and are very electron opaque. (See paper by Klotzel, J. Cell Sci. 15:379-401, 1974). Possible early fusion sites between the discoidal vesicles and the cytopharynx membrane are indicated by arrowheads. The elongated narrowed tips of the cilia extend into the nascent vacuole. Technical Details

Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol, and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. A print of the negative was scanned and processed in Photoshop. This image is best used for qualitative analysis.
